MD endonucleases work like traditional restriction enzymes, but, on the contrary, they cut only methylated DNA and may be used in epigenetic studies. <a href=\"#MD\">More details<\/a><\/p>\n<\/div>\n<div class=\"col-xs-3 bs-shortcode-col\">\n<p><mark class=\"bs-highlight bs-highlight-default\">GLAD-PCR assay<\/mark> &#8211; is a simple and inexpensive method for epigenetic studies of DNA preparations from clinical samples. It allows to detect even <mark class=\"bs-highlight bs-highlight-default\">several DNA molecules with R(5mC)GY site of interest<\/mark> in a presence of excessive amount of unmethylated DNA. Just 3 easy steps in one tube. <a href=\"#GLAD\">More details<\/a><\/p>\n<\/div>\n<div class=\"col-xs-3 bs-shortcode-col\">\n<p>Genomic DNA <mark class=\"bs-highlight bs-highlight-default\">fragmentation with GlaI<\/mark> and subsequent <mark class=\"bs-highlight bs-highlight-default\">NGS<\/mark> allow to construct a map of R(5mC)GY sites in analyzed genomes of patients and healthy donors. Comparison of these maps reveals R(5mC)GY sites &#8211; <mark class=\"bs-highlight bs-highlight-default\">potential epigenetic markers<\/mark> that can be used for diagnostics. <a href=\"#episeq\">More details<\/a><\/p>\n<\/div>\n<div class=\"col-xs-3 bs-shortcode-col\">\n<p><mark class=\"bs-highlight bs-highlight-default\">Light fraction of the blood cells<\/mark> is a good source of DNA for our PCR tests. A principal difference of new type of enzymes is that they cleave only methylated DNA. This property makes MD endonucleases a good instrument for epigenetic analysis.<\/p>\n<p>Now\u00a0 9 different MD DNA-endonucleases <a style=\"text-transform: initial;\" href=\"http:\/\/sibenzyme.com\/products\/m2_type\" target=\"_blank\" rel=\"noopener noreferrer\">are commercially available<\/a>. Recognition sites of MD enzymes differ in DNA structure and methylation patterns plus the enzymes may cleave the same recognition site in a different way.<\/p>\n<p>A special advantage of <a href=\"http:\/\/sibenzyme.com\/info627.php\" target=\"_blank\" rel=\"noopener noreferrer\">GlaI<\/a> recognition site R(5mC)GY &#8211; this site is the same as a product of DNA methylation with DNMT3 which is responsible for DNA methylation <em>de novo<\/em>. It is perfect for epigenetic DNA diagnostics in routine laboratory and clinical practice.<\/p>\n<p>It may be applied to <mark class=\"bs-highlight bs-highlight-default\">any source of DNA<\/mark>: tissues, biopsies, blood, plasma, serum, etc[\/vc_column_text][\/vc_column_inner][vc_column_inner width=&#8221;1\/3&#8243;][vc_column_text css=&#8221;.vc_custom_1576139259861{margin-bottom: 0px !important;border-bottom-width: 0px !important;padding-bottom: 0px !important;}&#8221;]The abnormal <em>de novo<\/em> DNA methylation is performed by DNMT3A and DNMT3B DNA methyltransferases. These enzymes recognize and methylate site 5\u2019-RCGY-3\u2019 with formation of 5\u2019-R(5mC)GY-3\u2019\/3\u2019YG(5mC)R-5\u2019[<a href=\"#pub\">1<\/a>]. Recently discovered site-specific methyl-directed DNA-endonuclease GlaI recognizes exactly this DNA sequence 5\u2019-R(5mC)\u2193GY-3\u2019\/3\u2019YG\u2193(5mC)R-5\u2019 and cleaves it as indicated
